Cell death&differ:Chinese scientists have discovered new strategies to suppress breast cancer stem cells

Recently, the International Journal of biology, cell death &differentiation, published a recent study by scientists from Canada and china. The researchers demonstrated that the activation of Toll-like receptor 3 by in vivo and in vitro experiments (TLR3) can promote breast cancer stem cells to tumor cells (CSC) transformation, have characteristics of stem cells, and the TLR3 Pro CSC conversion effect requires the simultaneous activation of beta catenin and NF- B signaling pathway can occur. This study provides a new strategy for inhibiting the production of CSC in cancer treatment.
 
The researchers pointed out that in the 2012 statistics, breast cancer is the second leading cause of death among women. Traditional therapies such as radiotherapy and chemotherapy can eliminate the tumor, but has few effects on malignant tumor cells still cannot afford to completely kill the role of these malignant cells can survive and self-renewal, further development of malignant tumors of this type of cells is called tumor stem cells. The study found that cancer stem cells are resistant to conventional therapies and can be converted to non neoplastic stem cells in some conditions. However, TLR3 has a higher expression in poor prognosis breast cancer patients, but the relationship between TLR3 and breast cancer cell transformation into tumor stem cells is still unclear.
 
Using cell cultures in vitro, the researchers found that activating TLR3 could promote breast cancer cells to produce tumor like stem cells, but TLR5,7,8 did not do the same. Through further study on the mechanism of activation of NF- alone, B signaling pathway and can cause breast cancer cells to tumor stem, while simultaneously activated catenin and NF- Beta Kappa B signal way, to promote the transformation of TLR3 mediated breast cancer cells to tumor stem cells. The researchers also found that the treatment of breast cancer cells with the use of cardamom can block beta catenin and NF- kappa B signaling pathways, leading to inhibition of TLR3 mediated CSC transformation.
 
In summary, the study found that TLR3 can also activate catenin and NF- Beta Kappa B signaling pathway promotes breast cancer cells to tumor stem cells, and found that cardamonin could block the effects of TLR3.inhibition of tumor cells to cancer stem cell transformation. This research provides a new strategy for inhibiting the production of CSC and improving the cure rate of cancer.
